Va estudiar saxofon alt i tenor, clarinet i violí abans de decidir-se pel saxofon baríton (ocasionalment tocava la flauta)

Fowlkes va passar la major part de la seva carrera primerenca a Nova York. Va tocar amb Tiny Bradshaw (1938-1944), Lionel Hampton (1944-1948) i Arnett Cobb (1948-1951).

Fowlkes es va unir a l'orquestra de Basie en 1953 i va romandre en ella fins a la seva mort; les principals interrupcions durant el seu temps amb Basie van ser les absències a causa de la gestió de la carrera de la seva esposa, la vocalista Wini Brown.
