Després d'estudiar clarinet, saxo i flauta, s'instal·la a Califòrnia, a començament dels anys 1940 i estudia amb Shorty Rogers. Toca, ja professionalment, amb Charlie Barnet (1947-1948), Art Mooney (1949) i, finalment, en la big band de Stan Kenton (1950-1951). Després, col·labora de forma continuada amb els Lighthouse All Stars d'Howard Rumsey, fins a 1956, convertint-se en un dels principals representants del West Coast jazz. Realitza vàries gires per Europa i Àfrica, amb Bob Cooper, introduint-se més tard en la fusió amb la bossa nova, de la mà de Laurindo Almeida.

En els anys 60 i 70, es dedica gairebé en exclusiva al treball d'estudi i a la música per a cinema, encara que grava alguns discos, per exemple, amb Ravi Shankar. Entre les seves col·laboracions d'estudi és particularment cèlebre el seu solo de flauta per a la cançó Califòrnia Dreamin' del quartet vocal The Mames & the Papes. A partir de 1974, represa la seva carrera com a instrumentista, formant una banda (L.A.Four) amb Almeida, Ray Brown i Shelly Manne. Alguns autors estimen que Shank va ser el flautista més important de la costa occidental, especialment considerant els seus reeixits duos amb el oboísta Bob Cooper. No obstant això, a partir dels anys 1980 abandona pràcticament aquest instrument, en favor del saxo alt, en un estil molt influenciat per Art Pepper i Zoot Sims, amb un control perfecte de la sonoritat, delicat i amb frasejo veloç.

Va morir d'una embòlia, als 82 anys, en Tucson (Arizona), on vivia, després d'acabar de gravar un nou àlbum.[
